Cover design  Photomicrographs from the brain of a 63-year-old man with hereditary frontotemporal dementia in a family with a mutation in exon 10 of the tau gene (P301L). Top panel shows classic flamed shaped neurofibrillary tangles in neurons of the CA1 sector of the hippocampus (modified Bielshowsky silver method). Bottom panel shows perinuclear `tangles' in neurons of the temporo-occipital gyrus [immunocytochemistry, anti-PHF-1 antibody (provided by S. Greenberg)] ABC^peroxidase^diaminobenzidine, haematoxylin counterstain, 1:800). The reasons for the variable distribution and morphology of these tau inclusions are unknown.
