Cover design  TUNEL, cytochrome c oxidase (COX) histochemistry and Hoechst 33258 staining on the same muscle biopsy sections of a patient with Kearns^Sayre syndrome, high percentage of single mitochondrial DNA deletion and severe deficiency of COX. (Right panels are double exposed photographs.) Top panels: TUNEL showed numerous myonuclei containing double stranded DNA fragments indicative of apoptosis. Most TUNEL-positive nuclei were present within COX-negative muscle fibres and some COX-negative areas in longitudinal fibres showed TUNEL labelling in all nuclei visible with Hoechst (fluorescent blue). Bottom panels: few TUNEL^positive nuclei (arrowheads) were also seen in COX-positive myofibres or in fibres with only mild reduction of COX staining.
